Borrelia spirochetes are the most common vector transmitted bacteria and divided into two different groups: Lyme disease and relapsing fever Borrelia. Complement is an important part of human immune defense, and pathogenic microbes need to evade it. One evasion mechanism is acquisition of the main host complement inhibitor, factor H (FH). We determined crystal structure of FH binding protein from Borrelia which enabled us to discover a structurally new immune evasion protein family found in both clades (LIAR-proteins) to which this proposal focuses on. Expression of proteins and mechanistic details how they help to evade immune defences of the host will be studied.Important is determination how LIAR proteins effect survival of Borrelia. New antigens will be identified to be used to improve diagnostic methods and as vaccine candidates. Studies will reveal more information on infection processes of Borrelia and describe new aspects of microbial evasion of complement.
