Project description
Fluorescence lifetime microscopy (FLIM) produces an image of the fluorescent probe distribution and the excited state lifetimes in a microscopic sample. As the probe lifetime depends on the environment of the probe FLIM can sense, for example, protein and DNA binding events as well as changes in pH and local viscosities. The project is focused on the application of FLIM to study extracellular vesicles (EVs) as innovative drug delivery systems in live cells. Using EVs, the natural carriers of different cargos between cells, for successful drug delivery has recently attracted great attention. However, the route of EVs in the cells is still poorly understood. First successful results of EV imaging in live cells performed in our group show the benefit of FLIM over conventional confocal microscopy. The project will provide further detailed FLIM study of EV trafficking in the cells using variety of fluorescent labels with different sensitivity (viscosity, pH).
