A key event in tumor progression is the establishment of an immunosuppressive milieu that prevents an effective attack by the immune system on the tumor. New cancer therapeutics targeting these mechanisms of immune evasion have emerged as a promising approach in multiple cancers. The aims of the study are to uncover the cell types and pathways that lead to immune evasion in ovarian cancer, and find novel ways to target these mechanisms with combination therapies. To this end, we are using a novel single-cell tumor tissue imaging technology, immune cell subpopulation gene expression analysis, modern artificial intelligence algorithms, and functional tumor-immune co-culture assays in a large set of >1500 ovarian cancer patients. With the proposed studies, we will discover the mechanisms of immune evasion in ovarian cancer, and design and implement an early phase clinical trial to select patients for novel combination therapies to improve the outcomes of ovarian cancer.
