The formation of memories of events is thought to be governed by the hippocampus together with the neocortex. It is unclear, however, how the saturation of this system is prevented to allow for new learning. We propose that a very brief event of intense cellular activation of the input part of the hippocampus during sleep, the dentate spike (DS), could provide the mechanism. We also suggest that new neurons born in the hippocampus throughout life are critical for DSs and thus in balancing synaptic modification. To test these assumptions, we will study 1) the contribution of DSs in learning tasks, 2) the functional connectivity of the brain related to DSs, 3) the effect of DSs on experimentally increased hippocampal synaptic strength, and 4) interactions of hippocampal cell types during DSs in both normal brains and in those without adult hippocampal neurogenesis. The results could reveal a new mechanism for retaining the plasticity of the hippocampo-neocortical system throughout life.
