Project description
This project sets out to shed light on initial pathological steps that lead to Alzheimer's disease (AD). Although macroscopically the two types of pathological changes in AD, amyloid plaques and neurofibrillary tangles, appear take place in different parts of the brain, a group of neurons in entorhinal cortex (in the medial temporal lobe) express both pathologies. These neurons express reelin protein, which is turn shares receptor with apoE, the main risk factor of AD. We aim to understand the interactions between reelin, Abeta and apoE by manipulating each using latest gene targeting techniques and viral vectors. In addition, neurons with Abeta accumulation tend become hyperactive but have compromised energy metabolism. We will assess if treatment of either alternation will slow down the spread of the disease process from its initial site.
