Project description
Current molecular targeted therapies intervene receptor tyrosine kinases (RTK) or specific Ras signaling arms but disappointingly, the redundancy of the kinase pathways has made it difficult to establish long lasting therapeutic effects. This situation has drawn new attention to non-redundant downstream proteins Myc and p53. The proposed research will determine whether Myc-dependent apoptosis can be reactivated in breast cancer by intervention of AMPK-dependent energy metabolism. The proposed research validates the Myc vulnerability based therapeutic strategy in novel preclinical breast cancer models. The drug studies will be performed in a factorial preclinical trial setting, using immunocompetent syngraft models of breast cancer and xenografts from patient-derived breast cancers. These studies are expected to shed light to Myc biology in breast cancer and offer for consideration the first clinically viable therapeutic Myc apoptosis reactivation strategy to fight breast cancer.
