Project description
Our recently gained expertise of 19F NMR spectroscopy is expanded to the characterization of bistable G-rich RNA regions that trigger pathological cascades in neurological diseases (including tumors). This knowledge is applied for the monitoring of small molecular ligand-RNA interaction. Of particular interest is how G-quadruplex-targeting ligands affect to plausible G-quadruplex/hairpin ratios (G-rich regions of oncogenic human NRAS, prion mRNA and related structures as models) and to the RNA-cognate protein interaction. The aim is to develop new drug candidates, but also to obtain new molecular level mechanistic aspects to the biological role of these G-rich RNA stretches. The latter part of the project focuses on the developing of novel neuron-targeting carrier molecules, the applicability of which to internalize small molecular ligands and oligonucleotide-based drugs to neurons in vitro and to target the central nervous system in vivo will be studied.
