Project description
Transcription factors (TF) are sequence-specific DNA-binding proteins. The aberrant overexpression of TF translocations are frequently observed in solid tumors including prostate cancer. Charting gene regulatory networks driven by the translocated TFs in cancer cells is a systematic way to the discovery of novel mechanisms and clinical markers for cancer risk prediction and therapy. Here we focus on the ETS fusions and cooperative TFs with significant roles in prostate cancer. One example is TMPRSS2-ERG or ETV fusions, which appear in over 60% of prostate cancer cases. We aim to uncover the mechanisms for these TFs and unique cancerous roles. Together with the data from genome-wide association studies, we aim to see how prostate cancer risk-associated SNPs alter TF DNA-binding to clinically important enhancers. We will validate key findings including risk SNPs, enhancers and genes that are potentially to be prognostic and diagnostic markers for prostate cancer risk prediction in men.
