Viruses are a significant cause of infectious diseases, and many viruses have a positive-strand RNA genome. These viruses drastically alter membranes of the host cells to replicate their genomes, and two types of membranous replication complexes have been recognized: invaginations and double-membrane vesicles. We study virus-induced membrane invaginations to define the factors required to form functional complexes. To this end, we developed a method to purify viral replication complexes from infected cells, and this method allowed us to obtain highly active complexes in large quantities. The purified replication complexes synthesized viral RNA in the test tube, and currently we are defining which host factors are essential. Collaboration between the University of Helsinki and Leiden University Medical Center has been an important part of the project. All in all, our data will be applicable for a number of important viruses, including pathogens, and will advance antiviral development
