Project description
Viruses are the leading cause of infectious diseases, and many pathogens are positive-strand RNA viruses. These viruses induce drastic membrane rearrangements in their host cells as they replicate in membrane-bound complexes. We are studying those complexes to define how viral replication is connected to the membrane deformation. Our aim is to establish assays which can be conducted in the test tube instead of cells. To this end, we will purify membranes containing viral proteins necessary for replication. Then, we will add RNA in the test tube to allow the membranes to form the replication complexes. This will enable us to dissect the key mechanisms of the replication and membrane alteration. This work will be carried out at the University of Helsinki in collaboration with Leiden University Medical Center and will be finished in three years. All in all, our data will be applicable for a number of important viruses, including pathogens, thus advancing development of antivirals.
